Methylene blue was synthesized in 1876 by a German chemist named Heinrich Caro. This is when it became a fabric dye used in the textile industry. The results of this study suggested that after a month of methylene e blue doses participants experienced a decrease in fear retention. Therefore, the study authors concluded, “Methylene Blue enhances memory and the retention of fear extinction”.
Methylene Blue helps with Bipolar Disorder
The use of methylene blue in the treatment of psychiatric conditions has been around since the early 1950s. Now we have evergrowing evidence that methylene blue has a positive effect on a well-known psychiatric condition called Bipolar Disorder. However, this study which was conducted at Dalhousie University in Halifax has only used methylene blue in concurrence with another anti-psychotic medicine called Lamotrigine. In this study subjects with a known case of Bipolar Disorder were given lamotrigine (Lamictal®) along with daily supplementation of either 195 mg or 15 mg of Methylene Blue. At the end of the study, the results concluded that the patients receiving a higher dose of methylene blue along with Lamotrigine had reduced depression and anxiety-induced symptoms. The patients believe it was so effective that they chose to carry on with the methylene blue supplementation even after the trial was over.
Methylene Blue nootropic dosage
Although recommended dosages differ from guideline to guideline, most agree on 0.5 – 4 mg/kg. So a 90 kg (200 lb.) body weight translates to 45 – 360 mg of Methylene Blue. This was deduced after a significant amount of animal and human trials.
However, your physician may recommend that you start at the lowest dose of 0.5 mg/kg and see which dosage suits you after seeing best and how your body and mind react to it.
In case you still want to start at high doses to amplify the effects, we recommend a dose of 10mg daily. This should be split up between the morning and night. A good route to consume it would be to take it with juice or milk.
Methylene Blue is a water-soluble compound which means when taken orally, you don’t need to take the supplement in relation to a meal which is a contrast compared to other Nootropics. Methylene blue also has a half-life of about 5 hours which is why it’s okay to take up to two doses a day
One thing to keep in mind whilst taking this supplement is that it may turn your urine blue, but it is nothing to worry about as it only happens if your daily dosage exceeds 500 mcg. But if it still bothers you, a hack to avoid blue urine is to take Methylene Blue with Ascorbic acid, commonly known as vitamin C.
Methylene blue supplementations are usually oral however it does come in an injectable form as well. But this can only be prescribed by a physician and is usually only used to treat diseases like malaria or as a part of a chemotherapy regime. . Alternatively, methylene blue can be taken in many forms including orally dissolved in liquid, powder in a capsule and topically applied on your skin.
What Are Liposomal Supplements?
Liposomal is a result of recent innovation in the health and technology industry. Liposomal supplements are unlike any other type of supplement previously manufactured. They have a special property that allows them to keep compounds with active ingredients fresh. It does this by surrounding the active ingredient particles in a layer of a fat-like substance known as phospholipids. This layer prevents the active ingredient from reacting with air or water on the outside and helps them remain potent for a long time. This kind of therapy has become a gateway to many delivery systems. For example, it can be used when active ingredients need to be kept protected until they reach a certain area of the body, this is called targeted therapy. Additionally, the layer of phospholipids is a safe and healthy replacement for binders, gelatin, or fillers used for the same purpose.
Liposomal Supplements Manufacturing Process
The manufacturing process of such an innovational delivery system is indeed very complicated. It involves a highly specific process that needs to be carried out meticulously. Additionally, it takes a long time to produce compared to ordinary supplements, and stabilizing the final product is a whole other task on its own. The process to develop liposomal-based pharmaceuticals has several major steps.
Material Dispensing, Dissolution, And Solution Sequencing
This step is often overlooked as it is the one that takes the longest however, it is justified in doing so as it requires great attention to detail. Usually, this phase requires between 5 and 10 rounds of buffers. Thes buffer can come in the form of phosphate salts, amino acids, sucrose, solvents, PEG, saline, and additional chemicals. The dispensing and dissolution phase is used to create a ‘homing’ layer between each layer of raw lipid. The steps to this phase usually include:1) emulsion preparation and solvent dissolution of the lipids.2) infusion of the API into the lipid and creation of the homing layer.3) stabilizing the final solution into a NaCl-based solution and removing all the impurities in the final formulation.This phase also requires numerous injections of water which may not sound important but it has to be carried out meticulously to reduce time later in the process. All the base materials are usually bought premade/procured to save time in manufacturing.
Liposomal Creation
The liposome is created by dissolving the raw lipids in ethanol (generally) and impacting them physically to create a liposome particle between 50 nm and 100 nm. The size range is dependent on the product’s target organ in the body and the physical biology required. The method always begins with a stable emulsion of water, solvents, and dissolved lipids. There are several methods of lipid transformation:1. Sonication: This is when ultrasound rays are used to irradiate the liquid emulsion created in the previous phase. The energy from these rays shatters the particles to form small spheres containing the active ingredient. However, this method only works with a small sample size.2. High-shear mixing/high-shear dispersion/homogenization: This method was first introduced by Szoka in 1983, This method uses careful control of energy levels, temperatures, pressures, and residence time. These parameters are then used to expose the emulsion previously created to energy. This requires stainless-steel vessels to contain the temperature, pressure, and byproduct recycling of the high-shear liquid/foam.3. Extrusion of the emulsion through a uniform pore-size polycarbonate membrane: This is a more commonly used technique for mass production. This method uses different qualities of filters to extract the dissolve lipid which is then streamed through the same filters multiple times. Of course, this whole process is carried out under specific time, temperature, pressure, and flow rates. This is the most popular method industrially because it requires lower energy and less machinery.4. Ultra-high pressure/ultra-high-shear micro fluidization: This method uses a pressurized mixing chamber that produces liposomal spheres quickly. However, this is not a popular method as it requires expensive high-pressure equipment with many safety hazards. Additionally, it required many support systems and skilled personnel to operate the equipment.5. Micro-chip/microchannel flow systems: This method uses innovational technology in the form of micro-sized chips to direct turbulent collision flow paths. This creates an environment that causes a lot of collisions which successfully makes liposomal spheres from the previously formed emulsions. This is popular since it doesn’t involve high energy or high volumes of solvent like the previous methods. This technique can be used in a lab but it can also be used at an industrial level.6. Jet injection systems: This technology involves a continuous flow path. Jet injection is used to create a precise and calculates steam that collides with a high-energy environment producing a continuous stream of liposomal spheres. This technique is advantageous industrially because it uses standard equipment that does not need specialized man force to handle it.
Liposomal Filtration
After the previous phase has been completed, we are left with a solution of various sizes and varieties of liposomes as well as byproducts and additives added during the liposome generation. This is why a filtration phase is necessary to sort the liposomes by their different varieties and exclude the byproducts. Usually, a selective filtration method is used by large ultrafiltration skids or tangential flow filtration systems. These methods use a selective membrane to filter out the liposomes. The latter of the two methods is more favorable as it is less time-consuming. It is compounded very well with microchip and jet injection methods in the liposomal generation phase as these methods produce smaller volumes of solvent and tangential flow filtration works best with smaller solvent volumes.
Liposomal Stabilization
After the filtration phase, we are usually left with a solution of a specific variety of liposomes in a solution of sodium chloride water. The solution is then further diluted to further eliminate any remaining byproducts. This phase also uses a variety of filters but usually, they are of the disposable kind. This is a tedious process as it usually takes a long time. After it is completed the liposome solution is refrigerated to prevent any form of degradation. Nowadays, products are always refrigerated between different phases of manufacturing.
Liposomal Loading
This phase of the manufacturing process is said to be the most chemically complex as we ll a significant step. This is the step that comprises the injection of the active ingredient into the carefully formed liposomes. Traditionally, the previously formed and filtered liposomes were heated at high temperatures to induce a chemical change in the pH so that the aqueous interior of the liposomes is exposed for the insertion of the active ingredient. The active ingredient easily penetrates the bilayer of phospholipids. This is of course carried out under extreme observation to ensure the potency of the end product. Once the active ingredient has melded with the layer the solution is then cooled down again to bring it to its former appearance of the liposomal structure, the only difference now is that the active ingredient is inside the liposomal spheres. After this is done some chemicals, like amino acids, sugars, PEG, and other chemicals used to control biophysical assimilation and stability, are used to attract the liposome to the target area of the body. These chemicals are used to count the outside of the liposomal spheres by rapidly heating and then cooling the solution causing pH changes for integration.
Final Product Stabilization
Depending on the outcome of the solution after the previous stage, it may need to go into further rounds of filtration to yet again purge the solution of impurities, byproducts, or other unreacted materials. This is a quick and easy step as it requires only the machinery previously used in the manufacturing process.
Aseptic Fill
Finally, the last step in this manufacturing process is sterilizing filtration and filling. Although the solution is almost the same density as water the fatty nature of the liposomes makes it a slow process when it comes to sterilization. There has been a lot of experimentation in this phase including increasing temperature, increasing pressures, increasing filtration ara=ea and so much more all in the favour of speeding up this phase. Unfortunately, none have been too successful as all of the above-mentioned tweaks tend to deshape or destroy the liposomes decreasing the yield.
Another part of the sterilization process includes ruining it through a bacteria challenge and bacteria retention test.
Liposomal Supplements Production Overview
As you can deduce the liposomal delivery mode includes many steps which require meticulous observation which makes the whole engineering process tricky and complicated. This makes the process arduous and costly since it requires skilled personnel and unique precise machinery. However, it is all worth it when realising the effectiveness of such a delivery system.
